Can I route my Steinberg Nuendo 2.0 or Cubase SX/SL 2.0 main outputs to multiple outputs on my audio interface?

You can go to VST connections, select 5.1 surround setup but just setup Left and Right. Right click on Add Child Bus, and you can assign that to the output.
